Vaccination against atherosclerosis: science or fiction?

چکیده

Atherosclerosis bears many similarities to inflammatory/autoimmune diseases like rheumatoid arthritis and multiple sclerosis (MS).1,2 Compelling data from experimental models show that such diseases may be treated by vaccination. Will it be possible to attack atherosclerotic cardiovascular disease with the same approach? Several studies have shown positive effects of immunization with antigenic LDL preparations,3–6 and a report in this issue of Circulation demonstrates a protective effect of oral and nasal immunization with another antigen, heat shock protein 65 (HSP65).7

Induction of cellular immunity against TIE2 by DNA vaccination attenuates atherosclerosis

TIE2 positive cels play a crucial role in processes that are potentialy involved in atherosclerosis, such as angiogenesis. Therefore, the specific deletion of TIE2 positive cels by means of DNA vaccination may affect atherosclerosis. Celular immunity against cels that overexpress TIE2 was established in LDLr -/mice by a novel oral DNA vaccination technique, in which an attenuated Salmonella typ...

Potential Benefits of Vaccinations

Introduction We live in an age of incredible technology, discovery and medicine. Smartphones, spaceships, and bionic eyes are no longer reserved for science fiction. Vaccinations are among the groundbreaking medical discoveries which “allow us to engage our adaptive immune systems to produce highly specific antibodies and immunological memory against a potential future infection” (Federman, 201...

Vaccination against VEGFR2 attenuates initiation and progression of atherosclerosis.

OBJECTIVE Vascular endothelial growth factor receptor 2 (VEGFR2)-overexpressing cells may form an interesting target for the treatment of atherosclerosis because of their involvement in processes that contribute to this disease, such as angiogenesis. METHODS AND RESULTS We vaccinated mice against VEGFR2 by an orally administered DNA vaccine, comprising a plasmid, encoding murine VEGFR2, carri...

Vaccination strategies in atherosclerosis.

The treatment of atherosclerosis is currently based on lipid lowering in combination with anti-inflammatory therapies that slow the progression of atherosclerosis. Still, we are not able to fully inhibit the formation or progression of atherosclerotic lesions. A very effective strategy in other disease pathologies is vaccination, in which the body is challenged with the culprit protein or micro...

DNA vaccination against VEGF receptor 2 reduces atherosclerosis in LDL receptor-deficient mice.

OBJECTIVE Similarities between neovascular ingrowth in atherosclerotic plaques and angiogenesis in tumors suggest that antiangiogenic factors that target tumor expansion may prove efficacious in the treatment of atherosclerosis. This study examined whether an oral DNA vaccine against the murine VEGF receptor 2 (Flk-1) with demonstrated antitumor effect through inhibition of pathological neovasc...
